چکیده
Cystic fibrosis (CF, OMIM 219700) is one of the most common severe hereditary diseases occurring in white populations. It is characterised by considerable heterogeneity in the mutational spectrum and a diverse phenotypic presentation, which justifies extensive studies on the correlation between the cystic fibrosis transmembrane regulator (CFTR) genotype and the CF phenotype. Genotype-phenotype data are available especially for common genotypes, but information remains scarce or lacking for genotypes of compound heterozygotes with a rare mutation and homozygotes for a rare mutation. Among rare CFTR gene mutations, 1811+1.6kbA>G is practically absent in CF patients from other parts of France, but it is not rare in patients genotyped in the south west of France, occurring in fourth place after F508del, G542X, and N1303K. Its frequency is 3.4% and higher than 5% with reference to the number of CF chromosomes with a south western geographical origin. The mutation 1811+1.6kbA>G, located in intron 11 of the CFTR gene, is (with the mutation 3849+10kbC>T) one of the only two splicing mutations of the CFTR gene that generates a new exon (1811+1.6kbA>G creates a donor site, 3849+10kbC>T an acceptor site). Furthermore, it was previously ranked by Chillon et al among class I mutations responsible for defective CFTR protein production and a severe CF phenotype. However, because it allows a reduced synthesis of normal protein, the mutation 1811+1.6kbA>G has also been reported in class V. 8 According to the recent classification proposed by Welsh et al, the mutation 1811+1.6kbA>G now belongs definitely to the “new” class I. The present report aims to study as well as possible the clinical severity of the mutation 1811+1.6kbA>G. The clinical data from 13 CF patients originating from the south west region of France and bearing 1811+1.6kbA>G (11 compound heterozygotes and two homozygotes) were collected.

Heterogeneity of phenotype in two cystic fibrosis patients homozygous for the CFTR exon 11 mutation G551D.
In the heterozygous state, the cystic fibrosis transmembrane conductance regulator (CFTR) exon 11 mutation G551D has been described as "severe," causing pancreatic insufficiency. Two cystic fibrosis (CF) patients homozygous for this mutation showed a mild rather than severe pancreatic phenotype and a variable pulmonary phenotype.
متن کاملNovel CFTR Mutations in Two Iranian Families with Severe Cystic Fibrosis
Background: Cystic fibrosis (CF) is a common autosomal recessive disorder that affects many body systems and is produced by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene. CF is also the most frequently inherited disorder in the West. The aim of this study was to detect the mutations in the CFTR gene in two Iranian families with CF. Methods: After DNA extractio...
متن کاملAnalysis of CFTR Gene Mutations in Children with Cystic Fibrosis, First Report from North-East of Iran
Objective(s): More than 1500 registered mutations in cystic fibrosis transmembrane regulator (CFTR) gene are responsible for dysfunction of an ion channel protein and a wide spectrum of clinical manifestations in patients with cystic fibrosis (CF). This study was performed to investigate the frequency of a number of well-known CFTR mutations in North Eastern Iranian CF patients. Material and...

OBJECTIVE To assess the concentration of faecal elastase-1 (EL-1) in pediatric patients with cystic fibrosis with mutation DeltaF508. METHODS Cross-sectional study with samples collected consecutively from 51 patients aged 4 months to 17 years old (mean 9.11±4.74); 32 (62.8%) patients were male. Clinical-demographic data were collected, as well as data on the type of mutation. Exocrine pancre...
متن کاملThirteen cystic fibrosis patients, 12 compound heterozygous and one homozygous for the missense mutation G85E: a pancreatic sufficiency/insufficiency mutation with variable clinical presentation.
To study the severity of mutation G85E, located in the first membrane spanning domain of the CFTR gene, we studied the clinical features of 13 Spanish patients with cystic fibrosis (CF) carrying this mutation. G85E accounts for about 1% of Spanish CF alleles. One patient was homozygous G85E/G85E and the rest were compound heterozygotes for G85E and other mutations (delta F508 nine patients, del...
